Hygromycin B is an aminoglycosidic antibiotic that inhibits protein synthesis in prokaryotes, by inducing the misreading of the m-RNA template, and eukaryotes.
It is used to select and maintain the phenotype of eukaryotic cells that are stably transfected with the E. coli hygromycin-resistance gene (hyg or hph). This antibiotic exhibits monophasic inhibition of polypeptide synthesis, suggesting that it binds to a single site on the ribosome. This action causes the mRNA sequence to be misread, resulting in protein synthesis inhibition. Hygromycin B is also an antiviral agent, which works as a standard section antibiotic in gene experiments.
